Chromosome walking is a technique to clone a gene (e.g., a disease gene) from its known closest markers. The closest linked marker (e.g., EST or a known gene) to the gene is used to probe a genomic library. A restriction fragment isolated from the end of the positive clones is used to reprobe the genomic library for overlapping clones. This process is repeated several times to walk across the chromosome and reach the gene of interest.
