iodine solution is used to test leaves for the presence of starch. You need to add the leaf to boiling alcoho(C2H5OH) for a few minutes (the boiling ethanol(C2H5OH)dissolves the chlorophyll and removes the green colour from the leaf - it turns white so it is easy to see the change in colour
this removes most of its colour.
In order to see the changes brought about by the iodine test , we have to remove the green colour. The green colour is given by the chlorophyll when we wash the leaf with alcohol ,we are actually removing chlorophyll . then only we will be able to observe that changes during iodine test
